Amorpha fruticosa L. distributs widely in China. The fruit, called "Zi Sui Huai " is a folk Chinese medicine in China showed the activity against the ulcer, eczema, burn-scald and so on. Although a lot of studies on chemistry and pharmacology of Amorpha fruticosa L. wre reported in other countris, there are not any research on chemistry , the anti-tumor activity and HPLC finger printer chromatogram in China. In order to exploit the plant rescoures and find most new antitumor compounds, the following studies were carried out.1. The 75% ethanol (FAF-75E) extract of the fruits of Amorpha fruticosa L. was suspended in water and partitioned successively with PE (FAF-P), EtOAC (FAF-A) and BuOH (FAF-B). The activity of every layer including the residues of aqueous (FAF-W) was evaluated by the same method. The bioactive parts of inhibitory effects on inflammation and pain were determined by the pharmacological tests. The fact was shown that the activity of FAF-P and FAF-A was more potent.2. Further invastigation on the chemical constituents of the extract of FAF-P and FAF-A by all kinds of chromatographic methods such as silica gel, preparative TLC and HPLC etc., 18 monomers have been isolated from the fruits of Amorpha fruticosa L., and 14 monomers including 1 stilbinoid, 11 flavonoids, 1 triterpene and 1 fatty alcohol, were identified by physico-chemical and spectral methods. They are 3,5-dihydroxy-4-geranyl-bibenzyl (1), 5,7-dihydro-8-geranyl-flavanone (2), 6a,12a-dehydrodeguelin (3), 6a,12a-dehydro-α-toxicarol (4), tephrosin (5), dehydrosermundone (6), 7,4' - dimethoxyisoflavone (7), 5-hydroxy-7,4' -dimethoxyiseflavone (8), 7,2' ,4' ,5' -tetromethoxyisoflavone (9), 2' -hydroxy-4,4' -dimethoxychalcone (10), 6a,12a-dehydroamorphin (11), kaempferol-3-glucoside-7-rhamnose(12), β- sitosterol (13) and n-triacontanol (14). Among them, compound 11 is a new compound, compound 6 is a new natural compound, Compound 1 is for the first time isolated in family Leguminosae, compound 8, 10, 12, 13, 14 are for the first time isolated in genus Amorpha, and compound 2, 3, 4, 5, 7, 9 are obtained from the fruits of Amorpha fruticosa L. grown in China for the first time.3. HPLC fingerprint chromatographic quality control standard of the the AF flavonoids, which are the effective parts of the fruit of Amorpha fruticosa L.was developed through 14 collected samples of differet origin in China. A reverse-phase HPLC method has been developed for the determination of 14 samples. Separations were achieved with a reversed-phase C18 column and the solvent system employed was methanol/acetonitrile/water/phosphoric acid 35:26:39:0.17 (v/v/v/v). The flow-rate was kept constant at 1.0ml/min with column temperature at 30°C with a UV detector setting at 295 run. The obtained HPLC fingerprint chromatogram is up to standard based on the results of methodology. Based on the results of HPLC fingerprint chromatogram, the intergral numerical value of the co-existing peak area (contents of AF flavonoids), similarty an the contentsof 3,5-dihydroxy-4-geranylbibenzyl, tephrosin and 7,2' , 4' , 5' -tetromethoxy-isoflavone were chosen as the quality control indices to the quality assessment of medicinal materials. The sample extraction condition such as extraction methods, extraction solvent, extraction time and purified method of flavonoids were optimized. The results showed that the contents of flavonoids were quite differet in different origins and collected time are better on September than other month. The contents of 3,5-dihydroxy-4-geranylbibenzyl, 7,2' ,4' ,5' -tetromethoxy-isoflavone and tephrosin in leaves of Amorpha fruticosa L. is less than that of fruit of Amorpha fruticosa L.. But it is very important because it is regeneration resources. The method of HPLC fingerprint chromatogram may be used to identify Amorpha fruticosa L. and its adulterants. It was proved that the technology of fingerprint analysis of TCMs is scientific, normative and effective method of the quality assessment of Chinese herbs.
