Objective: To determine the expression of PTCHmRNA and Smo mRNA in human pancreatic cancer cell lines, pancreatic carcinoma tissues and benign pancreatic disease tissues. To construct the recombinant adenovirus encoding human Smo antisense RNA. To investigate the role and its mechanism of Adenovirus-Mediated Smo Antisense RNA in inhibition of pancreatic cancer, in vitro and in pancreatic cancer xenografts in nude mice.Methods: RT-PCR was used to detect PTCH mRNA and Smo mRNA expression . construct recombinant adenovirus encoding human Smo antisense RNA with molecular biology technology. Cell proliferation were measured by MTT assays.and ~3H-TdR incorporation, cell cycle and cell apoptosis were detected respectively by FCM .The models of mice BxPC3 pancreatic cancer were used for in vivo antitumor test.Conclusions: there were over expressions of PTCH mRNA and Smo mRNA in human pancreatic cancer tissues and human pancreatic cancer cell lines.With molecular biology technology, we have successfully construct recombinant adenovirus encoding human Smo antisense RNA . Recombinant adenovirus encoding human Smo antisense RNA can remarkedly inhibit the growth and proliferation of pancreatic cancer cell in vitro and in vivo.
