Active portion were extracted from the Rhizoma Anemarrhenae, bio-assayed with pharmacological activities on rat cerebral ischemia model and five compounds were obtained from it. The active portion was named as the total timosaponins (TTS). The best extract technology and the purification conditions by absorption resins were optimized. The evaluation for pharmacological effect of TTS on several animal models indicated that the TTS was the active portion of Rhizoma Anemarrhenae, which could be used in the treatment of cerebral apoplexy. Microarray was conducted to investigate the protective mechanism of TTS on focal cerebral ischemia-reperfusion injury in rats. The results indicated that the protective effect of saponin B-â…¡ had several pathways. By means of HPLC-ELSD, the contents of four kinds of timosaponins were determined. The contents varied with the collected location and the further result showed that regions of cultivation, harvesting times and storage periods could affect the quality in a certain extent. Then the fingerprint analytical method was established. The samples from different resources were all of high similarity. Thus the HPLC-ELSD fingerprint method is repeatable and feasible and can be used qualityassessment of Rhizoma Anemarrhenae in combination with content assaying.
