RHIZOMA ANEMARRHENAE and RHIZOMA POLYGONI CUSPIDATI are TCM commonly used for clinlic treatment. As a newly developed quality control method for TCM, the chromatographic fingerprint can evaluate the overall quality of TCM. As a result, the method has been acceptable by lots of country in the world as the most effective way to control quality of TCM or nature products. In this paper, RHIZOMA ANEMARRHENAE and RHIZOMA POLYGONI CUSPIDATI were studied by HPLC and HPCE respectivly, the active components of which were determined also. Moreover, the evaluation of quality of the two herbs were carried on by chromatographic fingerprint method.Part 1. Study on the chromatographic fingerprint in quality control of RHIZOMA ANEMARRHENAE by HPLC.Firstly, neomangiferin and mangiferin in RHIZOMA ANEMARRHENAE from different sources were determined by HPLC. Extraction methods were evaluated for the extration efficiency of neomangiferin and mangiferin. In this experiment, three factors were studied by orthogonal design,including methanol concentration,time of sonication and times of extraction. The optimal extraction condition was obtained with 80% methanol, the extraction was repeated 3 times and each time for 20 minutes. Secondly, a standard chromatographic fingerprint was established, based on chromatography of 15 batches of RHIZOMA ANEMARRHENAE ,which obtained from different sources and in different growth period. And the standard fingerprint was used to assess the quality consistency of RHIZOMA ANEMARRHENAE from different sources. Good similarities with correlation coefficients higher than 0.9 were found in fingerprints between the herbs from different sources and the standard chromatographic fingerprint. Furthermore, the evaluation standards and influence factors on the similarity evaluation system for chromatographic fingerprint of TCM were discussed in-depth. Thirdly, the Heuristic Evolving Latent Projection(HELP) method was introduced in the study on chromatographic fingerprint method in RHIZOMA ANEMARRHENAE, which has been applied in the resolution of overlapping chromatographic peaks in RHIZOMA ANEMARRHENAE, and satisfactory results were obtained. It was shown that the combination of chemometric methods and modern analytical instruments will provide a new way for analysis of TCM. Part 2. Study on electrophoretic fingerprint in quality control of RHIZOMA POLYGONI CUSPIDATI by NACE.Firstly, Resveratrol-3-O-b-D-glucoside, Emodin and Rheochrysidin in RHIZOMA POLYGONI CUSPIDATI from different sources were determined by NACE. Extraction methods were evaluated for the extration efficiency of Resveratrol-3-O-b-D-glucoside, Emodin and Rheochrysidin.Secondly, based on electropherogram of 6 batches of reference RHIZOMA POLYGONI CUSPIDATI ,a standard electrophoretic fingerprint was established,with 12 common peaks in the fingerprint.Then the standard fingerprint was used to assess the quality consistency of RHIZOMA POLYGONI CUSPIDATI from 5 different sources.Good similarities with correlation coefficients higher than 0.9 were found in fingerprints between the raw material from different sources and the standard fingerprint.It was shown that the application of electrophoretic fingerprint method in quality control of RHIZOMA POLYGONI CUSPIDATI by NACE will provide more accurate evaluation standard of its overall quality. Thirdly, the basic mechanism of NACE was studied.The influence of some factors on EOF and electrophoresis mobilies was studied in capillary electrophoresis in methanol. It is the point to study the influence of BGE ionic strength(I) on electrophoresis mobilies,and it is shown that the mobility as a function of BGE ionic strength is in good agreement with the theory of Debye-Hückel-Onsager(DHO),extended by the ion size parameter as introduced by Falkenhagen and Pitts,indicating that the migration of the analyte ions in capillary electrophoresis in methanol can be predicted to some extend. ...
