We investigate the different phenotype frenquence of Rh blood groupand RhD (-) blood supplying in two regions,analyses the main reason of difficult blood crossmatchin pre-trasfusion testsand and study the number of the RhD antigen on different kind of RhD antigen phenotype.These can help understand the application range of mPEG modified RBC and establish the evaluation standard of RBC modification results. We try to camouflage the Rh blood antigen on RBC with varied MW mPEG, find the most suitable chemical among 7 kinds of mPEG derivatives and make the mPEG modification method which is hope to solve the rare Rh blood group phenotype blood supplying in China. We detect the in vitro markers for RBC structure and function which reflect if the mPEG modification can affect RBC structure and function.According to the detected results and the factors associated with RBC preservation, preliminary decide the mPEG modified RBC storage period. As basic study of the clinical safty of mPEG modified RBC transfused into human body, we focus on find if mPEG reacted with Rh antigen preotein is stable,if mPEG falls from RBC and the mPEG modification mechanism.We use FITC labeling mPEG-SPA modified RBC to transfuse into model anmimals mice and macaque to confirm the RBC survives and the safty for receiptor. At the same time, expore the most efficient animal models.At last, we try to use the modified RBC to crossmatch the serum from clinical samples which is difficult to crosmatch with general blood and compare which kind of blood crossmatch methods dose work for modified RBC and if the modified RBC can be transfusied for patient by the clinical standard.
