The recipe Wendan Tang has been widely used in clinical practice eversince it was developed.Based on Wendan Tang described in the 9th volume of Sanyin Jiyi Bingzheng Fanglun, doctors of the following generations have made up the prescription to satisfy clinical needs. This researching paper makes further systematic study on its medical effects on insomnia and its fuction mechanism on the ground of the results of the study of the past five yeas' literature related to Wendan Tang, taking insomnia as a breskthrough point.Objective: Investigate the sedative and sleeping effects of Wendan Tang in the 9th volume of Sanyin Jiyi Bingzheng Fanglun and the neuroendocrine-immune mechanism of treatment for insomnia. Methods: Detect the synergistically sedative and sleeping effects of the recipe and Nembutal sodium in mice by using routine pharmacological methods. Duplicate insomnia model of rats by means of PCPA abdominal injection. Study the neuroendocrine-immune of the recipe by observing the content of 5-HT, 5-HIAA, NE, IL-1β, TNF, GABA in insomnia rat brains and the immunohistochemical location of GABAA receptor and CCK8. Results: 1. The recipe,combined with Nembutal sodium, could promote sleep and decreas autonomic locomotion of mice; 2. Abdominal injection of PCPA deprives the nomal sleep of rats, at the same time, there appears decrease the contents of 5-HT and 5-HIAA and immunohistochemical location of GABAA receptor and CCK8 in the brains of insomnia rats. 3. After administration of the recipe, the decreased contents of 5-HT and 5-HIAA in the insomnia rats has increased to the monal level, while the content of NE decreased, compared with the normal group the difference is notable...
