Objective To study the main pharmacodynamics and mechnism of action of Anshen No. I on anti-insomnia, and to provide the theoretical foundation for developing new drug.Methods Methods of observing the direct sleeping, spontaneous and passive movement, coordinated motion, disappointment with tail hanging-up, the sleeping time produced by pentobarbital sodium, the quantity of sleeping animals caused by pentobarbital sodium in subthreshold hypnotic dose, the latent periods of sleeping produced by barbital sodium and effect on the convulsion induced by thiosemicarbazide after being given Anshen No. I orally to the mice were used to investigate the main pharmacodynamics of Anshen No. I on anti-insomnia.The variation of the content of L-NE, DA, 5-HT, 5-HIAA in brain was determined by RP-HPLC method with fluorescence detector to study the mechnism of anction of Anshen No. I on anti-insomnia.Results None of mice on every dose of Anshen No. I had a direct sleep. The middle and high dose of Anshen No. I could remarkably depress spontaneous and passive movements of mice, obviously prolong the sleeping time of mice produced by pentobarbital sodium and decurtate the latent periods of sleeping produced by barbital sodium, significantly prolong the latent periods of convulsed mice produced by thiosemicarbazide, decrease the number of death of convulsed mice and prolong the time of death. The high dose of Anshen No. I could decurtate the time of staying at the roll, prolong the time of immobility with tail hanging-up, increase the quantity of sleeping...
