Tumors that formerly were fatal now can be cured by cancer chemotherapy. However, many obstacles, such as excessive normal tissue toxicity, facing successful anticancer therapy and drug resistance may be either intrinsic or acquired. The multidrug resistance (MDR) is a unique phenome non-and characterized by tumor resistance to various structurally unrelated drugs. The mechanisms of MDR is still unclear, as to the former research there are about 4 mechanisms including the transportation of the cell membrane, the changes of cell and/or undercell structures, the genetic transformations and the targets changes. So the mechanism of MDR is complicated and not affirmed.Purpose: The aim of this study was to set up a feasible model to simulate the process and the presence of MDR and analysis the gene expression profiles by microarray technique.Methods: Part 1 , Induced MDR of the Tca8113 cell in vitro with CBDCA. The expression of MRP, MDR1 , GST- π , TOP Ⅱ were examined with RT-PCR and immunehitochernistry. Part 2, transplanted Tca8113 cell under the skin of nude mice to get the animal tumor model, then using CBDCA to induce MDR of the oral squamous cell carcinoma in the nude mice. Also examined the expressions of MRP , MDR1, GST-π , TOP Ⅱ.Part 3 , using microarray technique analysis the gene expression profiles of the transplanted tumors.Result: The expressions of MRP, MDR1 > GST- n , TOP II in Tca8113/CBDCA were higher than Tca813 whether in vitro or in vivo. Among the 14000 target genes, there were 719 genes whose expression levels differed between the Tca8113 and Tca8113/CBDCA in the transplanted tumors.Conclusion: cDNA microarray technique can simultaneously screen the different expression of genes from two different transplanted tumors. Further analysis of the obtained genes will help to understand the molecular mechanism of MDR.
