Fast haemostasis is the key for salvation of the wounded, so the development of effective haemostatic is the urgent need of war cure. Biological adhesive haemostatic mainly composed of fibrinogen and thrombin is the studied hot spot for Chinese and overseas scholars, now only a liquid fibrin sealant(FS) appears in market, FS is a kind of glue, but unfit for urgent haemostasis, now FS powder, dressing and so on become new developing haemostatics in fremdness, especially American army. The purpose of this study is to develop a new kind of fibrin haemostatic — lyophilized powder(fibrin haemostatic powder), which will be tested in clinic at the same time with or earlier than fremdness.Nitschmann fractions as the original material, purified methods of fibrinogen and thrombin were established by cold ethanol precipitation and ion-exchange chromatography on the principle of method simple, cost low, production easy, high recovery and purity. The stability of lyophilized thrombin was estimated with accelerated storage test, and the activity of thrombin would lose 10% after 161.8 days, 4.1 years, 6.2 years and lose 50% after 2.9 years, 26.7 years, 40.7 years kept at 25 ℃, 4℃ and 0℃ respectively. When coagulation time was guide line and less than 20 seconds, the optimized ratio of fibrinogen, thrombin and chloride calcium was estimated as 1 (clottable fibrinogen, mg) to 3.9-14.5 (thrombin activity, I.U.) to 0.21-0.60 (chloride calcium, mg) on the basis of camber regression analysis of SAS (statistical analysis system) software.The haemostatic effect of fibrin haemostatic powder (FHP) was evaluated according to three parameters (haemostatic time, blood loss and blood pressure decrease) on two species of the animals (rabbit and heparinized rat) and two models (partial liver resection and femoral artery incision). Treatment of the hemorrhagic animals was randomly divided into four groups: group I, negative control; group II, gauze; group III, thrombin; group IV, FHP. The result showed that FHP could stop blooding in three minutes on two models; on injury livers of rats, the blood loss in FHP group was less significantly than gauze group, but not than thrombin group, the pressure decrease in FHP group at the end of 30 min was less significantly than thrombin group; the blood loss in FHP group was significantly less than the other three groups and only the rats of FHP group were alive at the end of 30 min on femoral incision models, others all died.Fibrin haemostatic powder has simple purified technology, high constituent's purity and good haemostatic effect, will be a new haemostatic for war time and clinical use.
