Objective: To observe the effect of KNA on apoptosis of ovary granule cell in climacteric rat in order to explore the mechanism of KNA treating CS.Method: The animal model is to use natural senile rat to simulate the state of function descent in ovary. The target is to observe the different level of E2,FSH,LH,NO,MDA,SOD in serum and the apoptosis of granule cell in ovary and the correlated factors of apoptosis such as Fas, Bax, Caspase-3, Bcl-2, Bcl-2mRNA and BaxmRNA between the drug control groups and the climacteric group, the young compared group.Result:1.KNA could increase the E2 level while decrease FSH, LH in serum.2.KNA could rise the NO level in serum.3.KNA could decrease MDA level while increase SOD level in serum.4.KNA could inhibit the expression of BaxmRNA and theprotein expression of Bax, Fas, Caspase-3 while up-regulateBcl-2mRNA expression and the protein expression of Bcl-2.Conclusion:1.The animal model of CS could be reconstructed successfully.2.KNA could regulate the hormone level in serum of climacteric rats.3.KNA could up-regulate NO level thus it could affect the estrogen level in serum by the interaction of NO and estrogen.4.KNA could postpone senility course in some degree.
