Xueniaoting Experimental Study Of The Particles Purpura Nephritis, Cytokine And Apoptosis Regulatory Proteins

Objective: The goal of the dissertation was to observe the effect of Xue Niao Ting Granule on cell factor and mesangial cell apoptosis of experimental model rats. And to investigate the possible mechanisms of Xue Niao Ting Granule treatment on Henoch-Schonlein purpura nephritis (HSPN).Methods: The model of HSPN was established in rats by feeding Bovine Serum Albumin (BSA) and injecting BSA and Staphylococcus Enterotoxin B(SEB) into caudal vein. We copy the model rat of IgA nephritis to substitute HSPN. We observed the effect of Xue Niao Ting Granule on red blood cell and protein in urine and renal function of experimental rats. The level of Interleukin 1 and Interleukin 6 in blood and urine was detected by radioimmunoassay(RIA )and the expression of IL-6mRNA was described by in situ hybridization histochemistry (ISHH). We also studied the phenomenon of mesangial cell apoptosis by TUNEL and the expression of Bcl-2 and,Caspase-3 and Fas by immunohistochemistry SABC.Results: 1. The effect of high dose Xue Niao Ting, Xue Niao Ting plus Tripterygium wilfordii upon the lowering of blood in urine is statistically different compared with other treatment groups. (P<0.05). 2. Xue Niao Ting plus tripterygium wilfordii has biggest effect on the decrease of blood in urine. High dose Xue Niao Ting group is statistically different from lose dose of Xue Niao Ting and Bao Shen Kang groups. (P<0.05). 3. High dose Xue Niao Ting and Xue Niao Ting plus tripterygium wilfordii work best in decreasing the IL-1β and the IL-6 level in urine, which is statistically different from other treatment groups. However, its effect upon the decrease of IL-1β in blood and IL-6 in blood isn't statistically different from all treatment groups, which indicates IL-1β and IL-6 level in urine are the most sensitive cell detecting index. 4. To compare with Xue Niao Ting plus Tripterygium wilfordii group and Tripterygium wilfordii alone group, High dose Xue Niao Ting group isn't statistically different in decreasing the expression level of IL-6mRNA in ren, and lowering the expression of Bcl-2, as well as increasing...
