The Experimental Research On The Mechanism Of Sppan Acetic Ether Extrac On Immuno-Supression

Objectives: In this study ,throuth the expression both the cardial GrB that Sppan acetic ether extract to cardial allograft of SD rat and kidney TNF mRNA that MsPGN rat and the correlative indexs by means of microscope and RT-PCR, scope the immunate resist function and possible mechanism.Methods: Choose the improved Ono modle and replicate; the cardiac allograft model . After the harvest oprating ,we part these rats into three groups. groupA : model, groupB: CsA, and groupC: Sppan acetic ether extract; detect cardial GrB by means of microscope and RT-PCR .The rat modle of MsPGN caused by dulls seral bumin. we part forty rats into four groups, groupA: normal ,groupB : modle, groupC : Tripterygium wilfordii, groupD: Sppan acetic ether extract. After eight weeks,Observe the pathology changing of the cardial and kidney organation , the fuctions of the liver and kidney and kidney TNF mRNA by means of RT-PCR.Results: Sppan acetic ether extract can enlong the living time of the cardial allograft rat,refrain the expression of cardial GrB mRNA;reduce the pathology injure of cardiac ,the group CsA is better than Sppan acetic ether extrac; Sppan acetic ether extrac can resist the TNF mRNA expression of kidney,can inhibit the increasing of mesangium cell and the swelling of cellularstroma,decrease the deposition of immunocomplex in mesangium and glomer basement membrane,improve the role of proteinuria within 24 hours,and Sppan acetic ether extrac have no injure to liver and kidney.Conclusions: Sppan acetic ether extract can enlong the living time of the cardial allograft rat;refrain the expression of cardial GrB mRNA, it is possible one of the mechanisms that obstructing the cardial acute graft rejection.
