The origin and successive changes of the prescription, Pill of Four Miraculous Drugs (Si Shen Wan, 四神丸) was researched by document investigation, pharmacodynamic action and mechanisms of this prescription in inhibiting diarrhea was studied systematically and detailed by animal experiment.During the document investigation, we concluded that, Huatuo's Miraculous Prescription for Kid-Diarrhea is the first prescription for treating kid-diarrhea in extant record (recorded in Arcane Biography of Divine Doctor Huatuo, compiled by Sun Simiao, Tang dynasty). Afterwards, Chen Wenzhong of Song dynasty named this prescription as Pill of Four Miraculous Drugs (Si Shen Wan, 四神丸) in his contribution, Chen's elucidation on the presciptions for exanthema variolosum in pediatrics. From then on Pill of Four Miraculous Drugs (SI Shen Wan, 四神丸) became the representative prescription for treating kid-diarrhea and has been holding in esteem by numerous doctors.The animal experiments of anti-diarrhea and propelling rate of mouse's small intestine confirmed further that Pill of Four Miraculous Drugs (Si Shen Wan, 四神丸) played a role of firming intestien and inhibiting diarrhea. The experimental diarrhea caused by rhubarb, castor oil, folium sennae and pyridostigmine bromide in mice could be significantly antagonized by Pill of Four Miraculous Drugs.
