The first part: The light and the electron microscopechanges of brain in neonatal rat with HIBDObjective:To detect the pathological and infrastructure changes of brain inneonatal rat with hypoxic-ischemic brain damage (HIBD). Methods:The 7 day-old SD rats were subjected to the ligation of right carotid artery(ischemia),and then they were put into a box full with 8% oxygen and 92% nitrogen for 3 hours(hypoxia). The light microscope was used to observe the brain pathological changes and the electron microscope was used to detect the brain ultrastructural changes. Results:The changes of right side brain of rat had significantly pathological changes by 6-24h of HIBD and infarction of brain tissue had been observed by 72h group of HIBD compared with that in the control group. The number of neurons is significantly decreased in damagedgroups in the SD neonatal brain tissue within 6h ,at 12h and 24h, compared with the control groups P<0.05, and then gradually decreased. The expression of NgR protein was significantly increased in HIBD groups in the SD neonatal brain tissue within 6h, at 24h and 72h, compared with the control groups PO.05? and then gradually decreased. The expression change trends of NgR mRNA are identical with that of NgR protein. Conclusions:Above results suggest that the increasing of NgR mRNA and NgR protein may be correlated with the hypoxic-ischemic injury and may inhibit the neuron regeneration. The decrease in expression of NgR mRNA and protein may be correlated with the loss of nerve cell. The change trend of NgR and Nogo-A is identical.
