Objective To investigate the change of maternal serum interleukin-6 and interleukin-10 levels in preterm labor with chorioamnionitis , and their mRNA expression in placental tissues ,and the levels of IL-6 was interfered by IL-10 in human trophoblast cultured in vitro. To study the effect of IL-6 and IL-10 on monitoring preterm labor with chorioamnionitis , and the function of IL-10 on restricting preterm labor . Methods Partial placental tissues were detected after labor to be found out wheter chorioamnionitis or not in preterm labor and term labor in corresponding period , then preterm labor patients were divided into two groups by checking results: infectious group and noninfectious group,comparison of term labor.(1) Serum levels of IL-6 and IL-10 in all cases were measured by enzyme-linked immunosorbent assay. (2) The change of IL-6mRNA and IL-10mRNA were tested by reverse transcription polymerase chain reaction(RT-PCR). (3) Placentae of infectious preterm labor and non-infectious preterm labor were obtained after delivery and trophoblastswere be cultured in vitro, then challenged with 50ng/L of IL-10 and normal saline 1ml respectively in two groups, after the cells were incubated 72 hours , media and trophoblasts were collected, secretion of IL-6 in culture media was tested by ELISA and the change of IL-6mRNA in human trophoblasts was dectestded by RT-PCR. Results The levels of maternal serum IL-6 and IL-6mRNA of placentae in preterm labor with chorioamnionitis were significantly higher than that of without chorioamnionitis and term labor. (P<0.05).The levels of IL-10 and IL-10mRNA in preterm labor with chorioamnionitis was significantly lower than that of with chorioamnionitis and term labor, (P<0.05). the levels of IL-6 and IL-6mRNA were significantly higher in normal saline of infectious preterm labor than that of in IL-10,(P<0.05) and which were higher in normal saline of non-infectious preterm labor than in IL-10,(P>0.05). conclusion Maternal serum IL-6 and IL-10 can be a useful marker for detection of preterm labor with intrauterine infection early, IL-10 can restrain the expression of IL-6mRNA and the secretion of IL-6 in human trophoblasts . thus we think that IL-10 can control preterm labor caused by infection ,and plays an important role in maintaining normal pregnancy.
