To prohibit angiogenesis process in cancer can inhibit the growth of cancer. As one of endogenous antiangiogenic factor, angioarrestin plays an important role in endothelial cells proliferation, migration and blood vessel network formation. In the present thesis, the encoding gene for angioarrestin was cloned from human liver cDNA library by a PCR method;its MBP-tagged recombinant product was expressed and purified. Its polyclonal antibodies were obtained through immunization to Balb/c mice and its titer was about 1:1280 as determined by indirect ELISA and the antibody specificity was assessed by western blot. Stable transfected NCI-H460 cell line with angioarrestin and FD was also established. The cell growth curve was estimated, and the inhibition of the cell growth was obvious compared to the control. It indicated also that the cell transfected with FD domain appears with the characterics of higher adhesion and lower migration capability than those of control. The results showed that angioarrestin posses the activities of prohibit the cancer growth as well as anti-angiogenesis activities. Beside that, the mRNA expression pattern of angioarrestin after TNF α treatment was also observed in the present work.
