Adenoid cystic carcinoma(ACC) has a special characteristics of perineuralinvasion, so there is a great clinical significance to seek the mechanism of perineuralinvasion in ACC and to understand reasons for perineural invasion in ACC. In ourformer research, the research team presumed that many neurolin could combine withits corresponding acceptor which expressed by tumor cells,this might be thepathological histologic base of perineural invasion in ACC. It can be one of thereason for perineural invasion.Objective: To investigate the effect of SD rat sciatic nerve on proliferation,migration and adhesion of adenoid cystic carcinoma(ACC). And providetheoretical method for controlling the perineural invasion in ACC by researching on thepathological histologic base of perineural invasion in ACC. And to found solidbasis for diagnosis and treatment of ACC.Methods: 1. ACC-M cell line was co-cultured with conditioned medium andsciatic nerve culture medium for 24 48 and 72 hours. MTT was used to detect thein vitro proliferation of ACC-M.2. Sciatic nerve culture medium was setdownside in the Transwell room as a tendency factor, and co-cultured medium was act as control group. ACC-M cells were set on topside of the Transwell toinvestigate the effect of sciatic nerve on in vitro migration of ACC-M.3. ApplyMTT to detect the adhesion ratio of ACC-M cells which cultured by sciatic nerveculture medium, by which to research the effect of sciatic nerve on in vitroadhesion of ACC.Results :1. The proliferation of ACC-M cells incubated by sciatic nerve culturedmedium for 48,72 hours was significantly higher than that of othergroups(P<0.01),co-cultured medium had no effect on in vitro proliferation ofACC-M cells.2.Migration cell number of ACC-M cells in sciatic nerve groupwas significantly higher than that in the other two groups(P<0.05), andcompared to blank group, the ratio of ACC-M migration cell number was raisedby 51.5%, whereas co-cultured group was 8.1% higher, which had no differenceon statistics.3. After rebuild the artificial basilar membrane Matrigel, the ACC-Madhesion ratio of sciatic nerve group was significantly higher than that of theother two groups(P<0.05), whereas there was no difference between blank andco-cultured groups, by this we found that sciatic nerve culture medium couldimprove ACC-M cells in vitro adhesion to some degree.Conclusion :SD ratsciatic nerve culture medium could significantly improve the in vitroproliferation of ACC-M cell line, and it also raise the in vitro migration andadhesion of ACC-M cells to some degree. So we presume that during the ACCperineural invasion, nerve tissue may raise tumor cells'proliferation,migration andadhesion by its external secretion, and this might be the pathological histologic baseof perineural invasion in ACC...
