Objective: To study the expression of NgR knocking down by the siRNA and its effectiveness in promoting neurologic recovery and axon regeneration after an incomplete rat SCI. Methods: Two siRNAs were selected by using real-time RT-PCR as they had the greatest specific effect on NgR RNA replication. Contusion model of SCI in rats was used and the functional status of rats treated with siNgR and SAL post-injury was assessed using the combine behavioral score (CBS) 8 weeks later, after which the animals were killed for histologic studies. Results: The siRNAs could knock down the expression of NgR mRNA in cultured rat cortical and hippocampal cells; In rat SCI modle, the level of NgR mRNA and protein was knocked down also. siNgRs could promote neurologic functional recovery and axon regeneration of rats after SCI, which were demonstrated by CBS and Histological analysis and neural fibers staining. Conclusions: RNAi could knock down the expression of NgR and promote neurologic functional recovery and axon regeneration of rats after SCI.
