Effect Of The Du Meridian Acupoints With Acupuncture And Moxibustion Therapy On Nerve Regeneration Inhibit Protein Nogo And Nogo Receptor Complex In Vascular Dementia Rats

Objective: Compare and observe the effects of acupuncture, moxibustion,and acupuncture combined with moxibustion treatment respectively on vascular dementia rats in learning and memory function, as well as the effects of nerve regeneration inhibitory factor Nogo, Nogo receptors and its receptor complex expression.In terms of regulatory nerve regeneration inhibitory factors,the effects and possible mechanism of above treatment are investigated to provide experimental evidence and application reference for acupuncture and moxibustion on vascular dementia. Methods: 1.Rat model of VD was repeatedly induced lerebral ischemia and reperfused for two times, combined with intraperitoneal injection of sodium nitroprusside to establish the vascular dementia model. 2.120 SD rats were divided randomly into sham operation group(A),model group,(B),acupuncture group(C), moxibution group(D) and acupuncture and moxibution group(E).Acupoints were selected on the DU meridian of Bai Hui,Da Zhui,Feng Fu.At the same time, each group of rats were evalutated in learning and memory function. 3.Immunohistochemical technique was applied to Nogo(Nogo-A), receptor(NgR) and its receptor complex(LINGO-1) in three different time points(7d,14 d,21d) for each group. Positive cell expression of Nogo-A, NgR and LINGO-1 in each group of rats hippocampus CA1 region was analysed and compared; Western Blot detection technique was used to observe the expression change in Nogo-A,NgR and LINGO-1. Results: 1. Before treatment, there was no significant difference in learning and memory function among acupuncture group, moxibustion group, acupuncture and moxibustion group, model group. After treatment, about learning and memory function score, acu- puncture and moxibution group compared with the model group, there was a very significant difference(P<0.01);acupuncture group, moxibustion group compared with the model group,there was a very significant difference(P<0.01). Acupuncture and moxibustion group compared with acupuncture group, moxibustion group, there was significant difference(P<0.05). Acupuncture group compared with moxibustion group, there was a difference without statistical significance(P>0.05). 2 Expression of Nog- A, NgR, LINGO-1 positive cells Immunohistochemistry showed that compared with sham operation group, model group, the Nogo-A, NgR and LINGO-1 positive cells expression in the hippocampal CA1 at 7d began to increase; Nogo-A, LINGO-1 positive cells reached the peak at 14 d, compared with 7d,the positive cells expression of NgR was increased significantly; Nogo-A, LINGO-1 positive cells expression decreased,NgR expression reached peak;compared to the same period had significant difference(P<0.01).The positive cells of Nogo-A, LINGO-1, NgR expression were increased in the hippocampal CA1 at7 dfor acupuncture group, moxibustion group, acupuncture and moxibution group,compared with that of model group, the expression were decreased significant difference(P<0.01);The positive cells of Nogo-A, LINGO-1 expression reached the peak at 14 d, the positive expression of NgR was significantly increased compared with 7d, compared with the same period in model group, expression were reduced significant difference(P<0.01); positive expression of Nogo-A and LINGO-1 decreased,NgR expression reached peak at 21 d,compared with the same period in model group,There is extremely significant difference(P<0.01).The positive cells of Nogo-A, LINGO-1, NgR expression of acupuncture group, moxibustion group were higher than the acupuncture and moxinbution group at 7d, 14 d, 21 d, there was a significant difference(P<0.05). Between acupuncture group and moxibustion groups had no statistical significance(P>0.05). 3.Expression of Nogo-A, NgR, LINGO-1 protein Compared with the sham operation group, the expression of Nogo-A, NgR protein were increased significantly in hippocampus CA1 at 7d of model group.The expression of 14 d increased more significantly, Nogo-A, LINGO-1 protein expression quantity reached the peak at 14 d. the expression of Nogo-A, LINGO-1 protein were reduced, NgR protein expression reached peak at 21 d.The protein expression of acupuncture group, moxibustion group, acupuncture and moxibution group were lower than model group at each time point(P<0.05, P<0.05, P<0.01).Compared with acupuncture and moxibution group, the protein expression of the moxibution group were higher than the same period in acupuncture and moxibution group, there was a significant difference(P<0.05). Between the acupuncture group and moxibustion group protein expression was no difference(P>0.05). Conclusions: 1.Acupuncture, moxibustion Du Meridian treatment can improve vascular dementia rats learning and memory function. And acupuncture combined with moxibustion treatment is better than only acupuncture or moxibustion treatment. 2.Acupuncture, moxibustion treatment were able to inhibit the expression of Nogo-A,NgR and LINGO-1 protein,and acupuncture combined with moxibustion treatment inhibited the expression of Nogo-A, NgR and LINGO-1 protein was more significant. 3. The mechanism of acupuncture and moxibution treatment promote learning and memory functions of the vascular dementia rats may down-Regulated the expression of nerve regeneration inhibited factor(Nogo-A, NgR and LINGO-1 protein), and promote brain regeneration and repair.
