Objective: To find the tumor-associated genes in peripheral blood mononuclear cells(PBMC) from patients with pancreatic adenocarinoma by HUMAN GENOME U133 PLUS 2.0 ARRAY.Methods: PBMC from Patients with pancreatic adenocarinoma and normal control were envolved in this study. The total RNA of them was extracted. Hybridization with HUMAN GENOME U133 PLUS 2.0 ARRAY was performed. The image scanning and data analysis were performed subsequently. The partial results of hybridization were validated by real-time RT-PCR.Results: Expression profiling analysis of the 5 normal PBMC RNA samples and 5 pancreatic cancer PBMC RNA samples revealed that of 38500 genes on the HUMAN GENOME U133 PLUS 2. 0 ARRAY chip, 22000 probe set met the criteria for further analysis (at least one "present" in one sample). A fold change analysis and Student' s t test (two-tailed distribution;two-sample unequal variance) identified transcripts defined as differentially expressed between pancreatic cancer PBMCs and normal PBMCs. In total, transcript levels of 10 genes differed, on average, by 2-fold or greater between normal and RCC PBMCs, with P below 0.01 in a t test between the groups. MMP9, 0LFM4 and CCDC18 was validated by real time RT-PCR. And the results of real time RT-PCR showed the same expression trend as that in microarray. The expression level of 01FM4 in pancreatic cancer PBMC was significantly higher than that in normal PBMC(P<0.05).Conclusions: The tumor-associated genes in Peripheral Blood Mononuclear Cells(PBMC) from Patients with pancreatic adenocarinoma can be found by HUMAN GENOME U133 PLUS 2. 0 ARRAY. A group of tumor associated genes found in this study were likely to be the diagnostic marker and the potential marker of therapy.
