Objective:To investigate the relation between the expression of Eotaxin and recruitment of Eos as well as it's activition in allergic rhinitis and the effect of glucocorticosteroid. Then we explored the immunoregulation of histamine in allergic rhinitis throgh observing the effect of histamine on Eotaxin , IgE, Dcs and IL-12.Methods: (1) To establish allergic rhinitis animal model by OVA.Three groups was divided at random,namely normal control group, allergic rhinitis group without treatment, allergic rhinitis group treated with Dex. We detected the expression of Eotaxin and STAT6 protein by immumohistochemistry,semi-quantified Eotaxin mRNA by RT-PCR with 6-actin as internal contral,counted Eos in nasal mucosa throgh light-microscopy in high power field as the degree of Eos recruitment and detected ECP level in nasal lavage fluid by immunofluorescence as Eos activation extent;(2) We established allergic rhinitis animal model with experimental rats , which was divided into three groups: allergic rhinitis group without treatment,allergic rhinitis group treated with treated with histamine or HR1 antagonist;and set normal control group. To dectect Eotaxin , IgE and IL-12 level in serum or nasal lavage fluids by EIA;semi-quantified Eotaxin mRNA by RT-PCR, observed Eotaxin protein expression by immumohistochemistry and observeded DCs in nasal mucosa. Results: Compared with normal control group, expression of Eotaxin and STAT6 protein increased in allergic rhinitis group without treatment, ECP level and the number of Eos showed positive correlation with Eotaxin expression;After treated with histamine, Eotaxin expression and IgE prodction in allergic rhinitis rats increased and the number of S-100 positive DCs increased, but IL-12 level decreased .After treated with HR1 antagonist, opposite change occurred. Conclusion: Increased expression of Eotaxin in allergic rhinitis is an important factor resuting in recruitment and activivation of Eos, there are significant positive relation between them, glucocorticosteroid may play anti-inflammatin role thyough inhibiting the expression of Eotaxin.Histamine play an important immunoregulative role in allergic rhinitis.
