Objective: To study the effect of transplantation hRPE cell treatment of PD. Methods: hRPE cells from embryo were transplanted into the rat's striatum. Evaluating the therapeutic efficacy based on the change of rat's rotation behavior, PET scan and TH immunohistochemistry. The cells from eye bank were implanted in PD post-commissural putamen of 12 PD patients. Evaluating the therapeutic efficacy based on the change of CAPIT and PET scan. Results: PD rat model's rotational behavior and cellular morphology got much improved after cells transplantation. PET imaging show that hRPE cells could produce levodopa and had significant therapeutic efficacy in vivo. All 12 patients tolerated surgery well and no major adverse events occurred. And no obvious inflammatory response was seen. All patients have improvement in the primary outcome measure at 1 months post-operatively, 11 patients out to 3 months post-implant, the UPDRS score continued to improve. And PET showed increased dopamine release from 1 month post-implant. Conclusion: it is feasible to implant the hRPE cell expanded in vitro for long period in treating PD.
