The diseases induced by thrombosis, such as myocardial infarction and cerebral infarction, are becoming common and frequent over the world. The death rate caused by thrombosis has almost reached 40-50 percent. Recently, the study on the influential factors on thrombosis has made a great progress in basic and clinical research, which includes enhanced platelet adhesion and aggregation, raised blood viscosity and decreased activity in fibronolysis. However, thrombosis is a kinetic process, which intensively depends on local blood stream and wall status of blood vessel. In particularly, arterial thrombosis is based on blood vessel wall pathological changes, artery straitness and/or hyperkinesias, platelet activated by abnormal blood stream. The mechanism of connection and adherence in the network which involves receptor-counterreceptor or ligand interactions of platelet-leukocyte-endothelial cell crosstalk in thrombosis is a new focus in the research of anti-thrombus pharmacology and new drug. The methods of macro-hemorheology, cellular hemorheology and molecular hemorheology are employed in this study. The aim is to investigate the influence of plasma and blood viscosity changes on shear stress induced thrombosis and the relations between thrombosis and platelet, leukocyte, endothelial cell. In addition, the study is to find out the rules that the elements in the process of thrombus formation are changing with different shear stresses, time in cellular, molecular levels. Based on the above research, we investigate the effect of traditional Chinese herbal components, TMP and SAB, on shear stress induced thrombotic disorders. So we can find out the more effective combinative dosage, their advantages and possibility of synergistic. The aims of this study are to provide reasonable method for basic research on shear-related thrombosis, the experimental evidence for prevention and treatment of thrombus diseases, and the development of anti-thrombosis medicines.Part one: Investigations on Shear Stress Induced ThrombosisPlatelet-rich plasma (PRP) experiment: In this experiment, we discovered a number of facts: shear-induced platelet aggregation (SIPA) with changes in plasma viscosity, platelet micro-thrombus forming, the platelet aggregation rate varied with time, the relation between shear stress and aggregation rate, the relation between shear stress. In the experiment we recorded the change of platelet aggregation and plasma viscosity by...
