| Background:Coronary heart disease is affected by many factors as a pathological course, such as cigarette smoking, obesity, hyperlipidemia, hypertension, diabetes and so on . There exists a striking gender difference in coronary artery disease.Male sex is an independent risk factor for coronary heart disease after controlling other coronary heart disease risk factors. Existed reports show that the decrease of level of old man has a close relationship with the coronary heart disease growth and development,which show low level of testerone may be one of the independent risk factor for old man coronary heart disease.while hypertension, diabetes and obesity, as the independent risk factor,have sex hormon disorder.whether they have the same way to diease?how the sex hormon changed? whether they play a key role?what infection they play to blood pressure and blood now,the study about this way is little.usually,we think testerone play biologistic use through androgen receptor,while the expresstion and making function of androgen receptor is charged by gene.considering they play necessary role at different way,so we designed and maked this study.Object:(1) to study the relationship between TT,AR and gene polymorphism CAG.(2) to study their change and action at disease,and the relationship with disease,so to dicussion their role in the disease.Method:Takeing beijing wanshou road district 94 community as sample,take 7community from sample,we investigate old man 332 at all, 9 tumor are exclude,so the total man is 323 which include 104 healthy,hypertention 172,PAOD 44,stroke 62,di^bets 54,coronary disease 61,healthy aged from 60 to 85,mean 71.94±5.12, diabetes aged from 61 to 87,mean 73.03±6.26,healthy be diagnosised by standard designed by cnina medical associasion. first,to test human whole blood white cell AR by FCM.second,The test of AR gene polymomphism: takeing whole blood 0.1Ml,EDTAantisolidfy,use improved method to take patient whole blood WB DNA,which as sample,chose special out primer to enlarge androgen receptor gene CAG. Third,to test the level of sex hormon. Takeing whole vein blood 4ml,after be quite 1 hour,centrifugal 15 min,2500rpm,absorb supernatant,and be frozen at -80, ,and then be test at PLA hospital ,FT,DHEAS,SHBG be examine by ELISA through spnia Ttiturus automobil ELISA test,TT,E2,LH,FSH be test by chemical light way,use bayer Advia center to test.fourth,test blood blood automatic analyzer 747 test.Result:1.erect the method of testing human whole blood AR by FCM,which is a kind of way can test AR in cell and can test the quatity,only need very little blood.sensititivity and specialty are good,so,conforming to the need of development medical and clinical.2.provement human whole blood has AR.which show TT make affection to white cell through AR.3.the result of gene CAG polymorphism:the repeat of CAG from8 to 35.mean 22.7,the most repeat is 21(13.8%), 22(15.7%), 24(10.3%). (CAG)n<20take21%, (CAG)n>25 take 25.7%.4.the change of sex hormon make along with age. TT and age is nagative correlation,LH,FSH,SHBG is positive correlation with age.5.the relationship between sex hormon, AR, CAG and diabetes: TT is decreased clearly, AR make no sense,TT and AR act each other, and TT is nagative correlation with blood, CAG is no relationship with TT, Ar and diabetes.6. the relationship between sex hormon, AR, CAG ~nd obsity: TT is decreased, the main change is the HDL dowed, TT is the key factor.7. the relationship between sex hormon,AR,CAG and hypertention: TT is descend, SHBG, E2 is nagative correlation with BP.8.the repeat of CAG is no relationship with hypertention,diabetes, AR and sex hormon. but is positive correlation with BMI and coronary disease.Conclusion:1.AR is coactivitor2. CAG is gene polymornphism, no ralatiobship with disease, but has good correlation with BMI and coronary disease.3.CAG make no sense with sex hormon and AR,4.in sevev sex hormon,tt play a very key role, is very sensitive hormon, but absent from specifity,5.TT play very extensively , is a kind of protect factor.
