To explore the differential expression of toll-like receptors (TLRs) in human leukocytes and its significance in multiple myeloma(MM). First, we assayed serum samples from patients with multiple myeloma disease for quantitative immunolobulins. Based on fluorescent TaqMan-MGB technology, we establish real-time quantitative RT-PCR method to detect TLRsmRNA of peripheral blood mononuclear cells (PBMC) in 35 healthy donors and 50 MM patients . Second, we measured the positive rates of toll-like receptors in human lymphocytes, monocytes and polymorphonuclear leukocytes in patients with multiple myeloma disease and healthy donors by flow cytometry. The results showed that TLR1,2,4,6 mRNA level was significantly lower in patients with multiple myeloma than in control, but no significant difference was found between each type of multiple myeloma patients. TLR5 mRNA level was lower in patients with multiple myeloma than in health control , but no statistical significance was found. The positive rates of toll-like receptor 1,2,4,6 in lymphocytes and polymorphonuclear leukocytes were low in both groups, the positive rate of these toll-like receptors in monocytes in multiple myeloma group was significantly lower than that of controls, and the positive rate of the TLRs has no correlation with the typing of multiple myeloma.
