Objective:1: To study the role of airway inflammation on pulmonary arterial remodeling in rats with chronic bronchitis (CB) and emphysema during normoxic period.2: To study the characteristics of intra-acinar pulmonary arteries inflammation and its role on pulmonary arterial remodeling in rats with CB and emphysema during normoxic period. to study the ultrastructural changes of endothelial cells of intra-acinar pulmonary muscular arteries in rats with CB and emphysema during normoxic period.3: To investigate the role of vascular endothelial growth factor(VEGF) in remodeling of pulmonary arteries in rat with CB and emphysema during normoxic period.4: To evaluate the protective and therapeutic effects of erythromycin(EM) and antioxidant on intra-acinar pulmonary arteries inflammation and changes of endothelial cells of intra-acinar pulmonary muscular arteries and pulmonary arterial remodeling in rats with CB and emphysema.Methods:Male Wistar rats were randomly divided into normal control group, CB and emphysema group, EM-treatment group and NAC-treatment group. The rat model of CB and emphysema was established by intratracheal instillation of lipopolysaccharide(LPS) and daily exposured to cigarette smog. After eight weeks, artery blood gas analysis was observed, the pulmonary hemodynamics changes and cell counts in bronchoalveolar lavage fluid (BALF) were measured. The pathomorphological changes in the lung and the morphometric characteristics of the intra-acinar pulmonary muscule arteries were analyzed.
