Polygonum cuspidatum Sieb. et Zucc, a kind of perennial herb of Polygonaceae, widely distributes in the south areas from Qinling mountain of China. Its radix and rhizome are traditional crude drugs. Resveratrol and polydatin, which exist in P. cuspidatum, have been tested to have significant pharmaceutical effects and will become one of the most feasible medicines for preventing human cancer in the 21th century. At present, commercial extraction of resveratrol and polydatin are mainly to utilize the wild P. cuspidatum directly, which easily lead to natural resources exhausted. In order to realize the continuous utilization of this medicine plant resource, some systematic studies were performed about the biotechnology of P. cuspidatum, including tissue culture, genetic transformation, metabolism regulation, biotransformation and the screening tests of pharmaceutical activity. The main results were as follows:1 Established a system of rapid vegetative propagation of Polygonum cuspidatumA suitable system of rapid vegetative propagation of Polygonum cuspidatum was established by the method of plant tissue culture and a large number of plantlets were harvested in short time. Through a vast amount of screening tests, the suitable medium for bud formation was MS medium added TDZ 0.05 mg/L and NAA 0.5mg/L. After being treated with 0.1mg/L CPPU, 1.0mg/L NAA and 0.5% charcoal, the sterile seedlings grew quickly and propagated effectively. The 1/2 MS medium added 1.0mg/L NAA and 0.5% charcoal was successful for rapid rooting and strong seedlings. The bud explant of tissue culture seedlings of P. cuspidatum, as the initial material, was conducted to propagate rapidly in vitro and the intact plantlets could be gained...
