BK virus DNA was successfully detected in renal transplant recipients by real-time fluorescent quantitative PCR. A piece of nucleotide chain was constructed by a chemical way and was used to construction of external standards by gene cloning method; TaqMan probe was also applied in detection. This method is satisfactory in sensitivity, specificity, and repeatability. Cytology on BKV infection was carried out and was compared with PCR. Cytology has less sensitivity and specificity than real-time quantitative PCR. 63 renal transplant recipients were followed up for 8~10 months, the results showed that 3~4 months post-transplantation is a peak time for BKV shedding. Cold ischemia time is the risk factors for BKV infection. The viral load in urine samples from FK506 group is higher than from CsA group. But due to the lack of biopsy confirmed BK virus associatied nephropathy patients, a long-term perspective research on large samples are needed to further confirm the risk factors leading to BK virus infection and BK virus associatied nephropathy.
