Objective:To discuss the establishment and criterior of diagnosis of pressure-overload myocardial hypertrophy (POH), hypertrophy-reverse (POH-Re) and chronic congestive heart failure (HF) rat models. Methods:8-week-old Sprague-Dawley male rats were randomly divided into two groups: abdominal aortic constriction group (AAC) and sham group. AAC rats were performed abdominal aortic constriction to form POH, and age-matched rats were performed sham to serve as controls. After 12 weeks of surgery, all rats were performed echocardiography, and AAC rats whose LVmass/BW from echocardiographic detection increased were divided randomly into myocardial hypertrophy group (POH-NF, n=20) which were killed immediately, hypertrophy-reverse group (POH-Re, n=12) treated with Carvedilol (Roche, 0.5mg/kg/day) and Fosinopril (Squibb, 10mg/kg/day) for 8 weeks before being killed, and heart failure group (POH-F, n=29) on high-salt diet for 12 weeks before being killed. All the rats were detected their echocardiographic data and hemodynamic parameters through cadiocatheter via carotid artery before being killed.
