| Objective To evaluate the chemoresistance of primary head and neck squamous cell carcinoma to currently used chemotherapeutic agents in clinics including bleomycin, cisplatin + fluorouracil and paclitexal. Explore the expression of P-gp, MRP, LRP, GST- π ,Topo Ⅱ α and NF- κ B in primary squamous cell carcinoma, and the correlation between their expressions and chemoresistance respectively. Method 45 cases of head and neck primary squamous cell carcinomas were tested with MTT assay to evaluate their chemoresistance. The expressions of the 6 kinds of genes above were examined with immunohistochemical assay. Results There are 60.0%, 46.7% and 31.1% of the cases resistant to bleomycin, cisplatin + fluorouracil and paclitaxel respectively, and there are 20% of the cases resistant to all of the agents. The expression rates of P-gp, MRP, LRP, GST- π ,Topo Ⅱ α and NF- κ B in head and neck primary squamous cell carcinoma were 86.7%, 48.9%, 77.8%, 84.4%, 66.6%, 75.6% respectively. The correlation between the positive expression of P-gp and the chemoresistance to the three kinds of agents were significant, and the degrees of chemoresistance to cisplatin +fluorouracil among the different degrees of expression are significantly different.
