Cerebral ischemia is well known to be a common disease threaten to the old due to its high incidence, disability and mortality. In the past few decades, great efforts works has been done trying to clarify the pathphysiological process of cerebral ischemia, and many kinds of agents such as antagonists of calcium ion, blockers of glutamate receptor and free radical scavengers had been used in experimental cerebral ischemia. But few of them was shown to have a satisfying therapentic effect.3-n-Butylphthalide (NBP), a novel compound isolated from the seed of celery, has been shown to have protective effects against cerebral ischemia. It reduces the infarction volumn in rat brain after MCAO, ameliorates neurological deficits and energy metabolism, increases regional cerebral blood flow in striatum after MCAO, prevents the occurrence of stroke, and prolongates the life span after the onset of stroke in stroke prone spontaneously hypertensive rats (SHRsp). It appeared to be a new type of anti-cerebral ischemic drug. The present study was designed to further clarify the protective effects and the mechanisms of NBP on mitochondrial function, calcium overload and the expression of hsp70mRNA after cerebral ischemia in vivo and in vitro. The main results are as follows:Protective Effects of NBP on Mitochondrial Function1. ameliorating effect on the respiratory chain enzymes1. The activity of complex IV decreased sharply from 0.167 to 0.09 nmol·mg-1prot·min-1(P<0.01) after 1h-MCAO, the total content of mitochomdrial proteins also decreased from13.48 to 9.98 mg (P<0.01). Pretreatment with d1-NBP (5 mg·kg-1 or 10 mg·kg-1, ip.), theactivity of complex IV increased to 0.178 and 0.192 nmol·mg-1 prot·min-1 , respectively,and the total content of mitochondrial proteins also increased to 13.19 mg (5 mg·kg-1group).2.
