Essential hypertension is a common disease and a leading cause of cardiovascular morbidity and mortality. Its occurrence and development are results of complex interactions between a variety of genetic factors and environmental conditions. Genetics constitutes an important component of pathogenesis of hypertension. The renin angiotensin system(RAS) influences vascular tone, cardiovascular remodeling, and water homeostasis, RAS is also a prime suspect for harboring genetic determinants of hypertension. Therefore gene encoding component of RAS are attractive candidates for the investigation of genetic basis of hypertension. There is increasing evidence that variants of RAS gene are associated with hypertension in Western population, but remains poorly understood in Chinese han people. The purpose of this study is to observe the polymorphism of the RAS genes in Chinese Han People, and evaluate the genetic linking to hypertension. The polymorphisms of RAS gene related to Chinese han hypertensive were studied by molecular and genetic techniques, PCR, PCR-SSCP, Amp-FLP, direct sequencing of PCR products as well as subclone for sequencing. Pedigrees and individuals of Chinese Han people including 121 hypertensives and 95 normotensives were selected. Their age and sex were matched. All participants were in agreement with criteria. The studies were performed as following:1. ACE gene I/D polymorphism in Chinese han people In the hypertensive group, SBP/DBP(mean ± SD, mmHg) was 158 ± 17/98 ± 11, and in the control group, 114 ± 13/76 ± 7. No significant differences were observed in age, sex, BMI, Ch, TG, HDL-C, LDL-C,...
