Ischemic cerebravascular disease is a kind of common disease. It is quite harmful to human beings because of the high incidence, disability and mortality of this disease. It is well known that the causes of death and disability of patients is closely related to brain damage. In order to further study and elucidate the pathophysiological mechanisms of delayed brain damage after cerebral ischemia, and try to find a practical and efficient therapeutic method, a great deal of investigation has been conducted in recent three decades. However, the therapy of cerebral ischemia is still a big difficulty although a variety of anticerebral-ischemic drugs have been used in clinic.3-n-Butylphthalide(NBP), a novel compound isolated from the seed of celery, is an anticerebral-ischemic drug which has been shown to protect the cerebral ischemic damage, improve brain energy metabolism, reduce area of cerebral infarct and attenuate neurologic deficits in middle cerebral artery occlusion(MCAO) rats, and prevent the occurrence of stroke and prolong the life span after the onset of stroke in stroke prone spontaneously hypertensive rats(SHRsp). The present study was designed to further clarify the brain protective effects and mechanisms of actions of NBP by using different experimental methods (such as physiology, biochemistry, radio-immunology, cell culture, electro-microscope observation, high performance liquid chromatography(HPLC) and molecularbiology techniques etc.). The main results are as follows:(1) The protective effects of 1-3-n-butylphthalide(l-NBP) and d-3-n-butylphthalide(d-NBP) on 20 mM KC1- or 30 μ M NMDA- or hypoxia/...
