It is a desire for scientist to develope a new drug with high potency and low toxicity. The relationships between drugs' optical activities and their biological activities and toxicities have been found by the practice of many years. To develop an optical activity compound has been considered as a new strategy.The anticerebral ischemia effect and its mechanism of action of raceme ABP [3-n-butyl-6-aminophthalide, ( ± )ABP], a synthetic derivative of 3-n-butylpathalide (Ag-1),have been studied for several years in our laboratory on the levels of general, molecule and gene. Now, raceme ABP has been resoluted into levorotatory[(-)ABP] and dextorotation[(+)ABP]. The effects of chirality ABP on cerebral ischemia and its mechanisms of action were carried out as follows:( 1 ) Studied and compared the pharmacological actions of (+)ABP and (-)ABP onthe level of general, molecule and gene.( 2 ) Studied the effects of (+)ABP and (-)ABP on NO and NOS activity in primary cultured cell and global cerebral ischemia—reperfusion.( 3 ) Studied the effects of (+)ABP and (-)ABP on the apoptosis induced by cerebral ischemia—reperfusion.Study on the pharmacological actions of (+)ABP and (-)ABP on the level of general, molecule and gene1. In our previous research in vivo, the effect of ( ± )ABP was observed when it was administrated before cerebral ischemia. This study was designed to see if ABP could produce cerebral protective effect administrated after cerebral ischemia.
