The human eccrine sweat gland is an important skin accessory involved in secretion, excretion and thermoregulation . To separate and culture eccrine sweat gland epithelial cells in vitro would be of value in the study of its structures and functions further, in the understanding of fluid secretory mechanisms and in the elucidation of epithelial ion transport procedures and the mechanism of tubulogenesis. Furthermore, culturing the eccrine sweat gland cells in vitro makes it possible to fabricate the tissue engineering skin with accessories. At present, there are a few reports on sweat gland epithelial cells cultivation , which are only limited to tissue explant cultures and primary cultures. The cultured cells were impure and it took a long time to reach confluency. It is necessary to develop a more effective culture technique for isolating and culturing human eccrine sweat gland epithelial cells and try to reconstruct sweat glands in vitro.Therefore, this study was to establish a new and effective method to culture human eccrine sweat gland epithelial cells, to investigate the biological characters of the cells and possibility to reconstruct the sweat glands in vitro. Cell culture, immunohistochemistry, histopathology, westernblot and CLSM were used in this study. The methods, results and conclusions are shown as follows:Methods: 1.
