Background: Allergic atopic disorders, such as rhinitis, asthma, and atopic dermatitis, are the result of a systemic inflammatory reaction triggered by type 2 T helper (Th2) cell-mediated immune responses against 'innocuous' antigens (allergens) of complex genetic and environmental origin. A number of epidemiological studies have suggested that the increase in the prevalence of allergic disorders that has occurred over the past few decades. The initial interpretation proposed a missing immune deviation of allergen-specific responses from a Th2 to a type 1 Th (Th1) profile. However, in the face of discrepancies that have come to light from studies involving autoimmune diseases and helminth infections, it is important to reevaluate the immunological basis of the hygiene hypothesis. Increasing evidences has suggested asthma is an immune toleranic deficiency disease. More recently, the role of mature dendritic cells (DCs) and regulatory T cells (Tregs) has been emphasized. Due to the greatly increase of mature dendritic cells in bronchialalveolar, and the defeciency of regulatory T cells, type 2 T helper responses are enhanced when the atopic indivals exposured to allergens. The allergen-specific immunotherapy (SIT) represents the only curative approach for allergic asthma. DNA vaccines encoding the genes of allergens are new methods for specific immunotherapy, and are still in developing. DNA vaccines delivered by dendritic cells are more efficiency than plasmid DNA vaccines.Objective: To investigat the effect and efficacy of the dendritc cells transfected with genes of major allergens of house dust mite, Der p1 and Der p2, on the induction of immune tolerance in a mouse model of asthma sensitized and challenged with house dust mite extracts.Materials and methods: 1.
