Experimental Study Of Nanoparticles As An Adjuvant Vaccine Immunotherapy Of House Dust Mite

Objective: To build asthma murine model with Dermatophagoides pteronyssinus. To compare the curative effects of immunotherapy with Dermatophagoides pteronyssinus allergenic extract, recombinat Der p 2(rDer p 2) and nanoparticles encapsulated rDer p 2. To offer a new thread of immunotherapy to allergic diseases.Methods: Firstly, to prepare encapsulated rDer p 2 nanoparticles. Secondly, divide the BALB/c mice randomly into five groups. There are 8 mice in each group. They are: A. negative control, B. positive control I, C. allergenic extracts, D. rDer p 2, E. nanoparticles. On day 0,7, 14, mice were sensitized by i.p. injection of 100 μ l mixture of allergenic extracts and Al(OH)₃ except group A. During the day 28 to 30, mice of group C, D, E were i.p. injection of 100 μ l allergenic extracts, 100 μ l rDer p 2, 100 μ l encapsulated rDer p 2 nanoparticles respectively. During the day 44 to 49, mice of group B, C, D, E were i.n. given an challenge of allergenic extracts. Group A was treated by saline always. On day 51, all of the mice were sacrificed and their tissues were removed for study. Assays for IgE, IgG1, IgG2a in serum, IL-4, IFN-γ in BALF and SCCS were performed by ELISA. Total cell number and assoeted cell number in BALF sample were determined. The lungs kept in 10% formalin were made into slices stained by hematoxylin and eosin in order to observe the pathologic changes in lungs.
