Proteolysis processes are necessary for normal physiological functions in the body, including normal blood vessel maintenance, new vessel formation (angiogenesis), clot formation and dissolution, bone remodeling and ovulation. The cancer cells for their growth and spread also use the same enzyme system for the above roles. These enzymes produced by the cells can degrade the basement membrane and extracellular matrix (ECM) .The ECM degrading proteinases can be divided into three classes: metalloproteinases, cysteine proteinases, and serine proteinases. The uPA system of serine proteinases plays an important role in this process.uPA/uPAR are important in the uPA system, many articles had reported that there is no expression of uPA/uPAR in normal skin, but the expression is very high in the tissue of tumor and wound. Little is done about the expression of uPA/uPAR on the morphogenesis and the function on wound healing.Therefore, this research was designed to study the expression of uPA in the skin and hair follicle in the human fetus morphogenesis and the function of uPAR on wound healing from four aspects: firstly, the expression of uPA in the skin and hair follicle in human fetus morphogenesis; secondly, the Pam 212 cell were transfected by uPA ATF and pEGFPC1 recombinant plasmid or mock plasmid, named as Pam 212-pEGFPC1-uPA ATF and mock-Pam 212 respectively, after transfection, cell growth, invasion were observed; thirdly, the Pam 212 cell were transfected by uPAR and pEGFPC1 recombinant plasmid or mock plasmid, named as Pam 212 -pEGFPC1-uPAR and mock-Pam 212 respectively, after transfection, cell growth, invasion were observed; at last, the recombinant plasmid of pEGFPC1-uPA ATF, pEGFPC1-uPAR and mock pEGFPC1 were injected intradermally to the wound of mice, and electroporated at the site, the expression and the function of the recombinant plasmid were observed.The main results are listed following:1.
