Developing more effective antitumor vaccines has been one of the major goals of tumor immunotherapy. Previous studies of our department have confirmed in murine model that immunization with elemene combo-tumor cell vaccine could induce potent active immunoprotective antitumor effects, immunization with HSP70-peptide complex extracted from elemene combo-tumor cell vaccine could induce potent active specific immunoprotective antitumor effects. These results suggested that HSP70-peptide complex played a very important role in the course of immunoprotective antitumor effects induced by elemene combo-tumor cell vaccine.Increasing evidence suggested that dendritic cells (DCs) are professional antigen presenting cells that can prime adaptive T-cell immunity. Heat shock proteins (HSPs) are endogenous molecules that activate DCs. The ability of HSPs to potentially bind to the whole cellular peptide repertoire makes them attractive vectors for tumor vaccines.Based on these previous work , the present investigation examines the antitumor effects of DCs pulsed with HSP70-bcg and HSP70-ec-tcv on Hca-F and L929 tumor cells in vitro, the role of Toll-like Receptor 4 (TLR4) signal pathway in DCs activated by HSP70-bcg and HSP70-ec-tcv , the antitumor effects, in vitro and in vivo, of DCs activated with tumor antigens of Hca-F EC-TCV (Hta) and HSP70-bcg complex (HTA-HSP70-bcg) , reconstituted in vitro. The main results are as follows:I . Dendritic cells pulsed with HSP70-Bcg and HSP70-Ec-tcv1. We separated and purified HSP70.peptide complexes from Hca-F EC-TCV cells and BCG.
