Objective To investigate whether allogeneic dendritic cells pulsed with heat-shock tumor cell lysates(hTCL) could induce antitumor responses Against Lung Cancer mediated by T-cell.Methods hTCL or tomor cell lysates (TCL) extracted from NSCLC patients were used to pulse DCs from patients or Allogeneic donors, and with suitable condition, peripheral blood mononnclear cells(PBMCs) were elicited to response against lung cancer, FACS were used to monitor the expression of the costimulatory molecules on the surface of DCs, inducible hsp-70 in hTCL were detected by Western-blot, hspenzyme-linked immunospet (elispot) and lactate dehydrogenase release(LDH)assay were used to monitor the T cell population released IFN-yand the specific killing activity.Results Hsp-70 in the cancer cells can be induced by 41.5℃heating。Allogeneic immature dendritic cells showed full ability to be enhanced with the expression of CD86 molecules on the surface and the secretion of IL-12, which indicated that the hTCL promoted the maturation of DCs. And the corresponding T cells displayed strong capability of releasing IFN-yand killing tumor cells line of A549。conclusion after mixed culture with PBMCs from lung cancer patients in vitro, Allogeneic Dendritic Cells pulsed with hTCL, as the immunogen, can stimulate and amplificate lung cancer reactive T-cell, and may be used as the tumour vaccine with high performance.
