Chemokine is a kind of cytokines,which can been induced.It is a kind of small polypeptide molecular and result from many cells.They include four class:CXC;CC;CX3C;C.SDF-1a (CXCL12)belong to CXC family.There is 99% homology between human SDF-1a and murine SDF-1a.The difference with other chemokines is that SDF-la is secreted continuously from matrix cells and do not inducing express resulting from inflammation. CXCR4 is a solely receptor of SDF-1a.It not only express on cell surface,but also express in cytoplasm. CXCR4 widely express in immunity system and central nerve system.CXCR4 expression has recently been characterized in several cancer cell types and is thought to play a pivotal role in directing the migration of metastasizing tumor cells to CXCL12-rich tissues. A.Muller et al(2001)and R.J.Phillips(2003)respectively reported,CXCL12, which is highly expressed in the organs to which breast cancer and non-small cell lung carcinoma (NSCLC)preferentially metastasize, has been shown to promote cancer cell migration This ability to promote cancer cell migration can be inhibited with anti-CXCR4 antibodies. Similarly,H.Geminder(2001)et al. showed that conditioned media from bone marrow stromal cells can support neuroblastoma cell migration through a CXCR4-mediated mechanism.D.Uchida et al. ( 2003 ) demonstrated that CXCR4 mRNA expression is higher in metastatic lymph nodes than in primary HNSCC (Head and neck squamous cell carcinoma) tissues. Furthermore, it was shown that CXCL12 in lymph node extracts mediates HNSCC migration in vitro. Thus, the results shown herein support the hypothesis that the CXCL12-CXCR4 axis plays a key role in HNSCC metastasis to lymph nodes.The murine hepatocarcinoma cell line (Hca-F) and its syngeneic cell line(Hca-P)were isolated by professor Ling Maoying from distant hybridization Km murine ascetic hepatocarcinoma cell line H22,and both of them can specially metastasize to lymph node. When they are injected into 615 mouse's heel,Hca-F cell's lymph node metastasis rate is higher than 80%,while Hca-P cell's is lower than...
