Objective:To investigate the expression and clinical significance of the chemokine receptor-4(CXCR4) and matrix metalloprotease-13(MMP-13) and the relation among the expression of the two factors with tumor invasion and metastasis in laryngeal squamous cell carcinoma(LSCC).Methods:CXCR4and MMP-13gene protein in70cases of laryngeal carcinoma and10cases of normal tissue around the cancer were detected by immunohistochemistry.CRCR4and MMP-13mRNA in30cases of laryngeal carcinoma and10cases of normal tissue around the cancer were examined by reverse transcription polymerase chain reaction(RT-PCR).To study the relation between the two factors with clinicopathological parameters and tumor metastasis by two experiments, the laryngeal carcinoma were devided by age, sex, clinical stage, depth of invasion and lymph node metastasis.Results:Immunohistochemistry and RT-PCR both show:The positive expression rate of CXCR4and MMP-13in tumor samples was significantly higher than that in normal ones, and the differerces were statistically significant(P<0.05), both significantly higher than normal tissue;In tumor samples, the expression of CXCR4were not associated with age and sex(P>0.05), while it were higher in clinical stage, depth of invasion and lymph node metastasis(P<0.05). Over-expression of MMP-13was significantly correlated with depth of invasion and lymph node metastasis (P<0.05).The expression of MMP-13were not associated with age, sex and clinical stage. The expression of protein and mRNA were at the same level.The expression level of CXCR4and MMP-13in the laryngeal squamous carcinoma tissue positively correlated.Conclusion:1. CXCR4and MMP-13may be involved in the laryngeal squamous carcinoma, and may play an important role in the invasion and metastasis of laryngeal squamous carcinoma.2.CXCR4and MMP-13play a cooperative role in the process of laryngeal squamous carcinoma, which suggested that they would become a biologic indexes to prognosis of laryngeal carcinoma.3.The overexpression of CXCR4was related to MMP-13, which suggested that they would be up each other.
