Salivary gland mucoepidermoid carcinoma is the most frequently encountered malignance in salivary gland. The high-grade mucoepidermoid carcinoma is characterized by high metastasis and poor prognosis. The metastasis is the main reason of death. Treatment of the high-grade mucoepidermoid carcinoma is surgery with adjunct chemotherapy or radiotherapy. So far, there is still no effective treatment for mucoepidmoid carcinoma and the 5-year survival rate is low. Therefore, it is necessary to develop new stratagem for the treatment.RNA interference (RNAi) provides a powerful method for gene silencing in eukaryotic cells, in which the recognition of double-stranded RNA (dsRNA) ultimately leads to post-transcriptional suppression of gene expression. It has become the most powerful technology in biology. The use of RNAi for genetic-based therapies is widely studied, especially in viral infection, cancer, and inherited genetic disorders. DNA vectors constructed to mediate RNAi by expressing short hairpin RNA (shRNA) are a newly established technique that can produce stable, long-term, and highly specific gene silencing. RNAi technique may become a new method for gene therapy.Transforming growth factor-β (TGF-β1) is a multifunctional peptide that elicits numerous cellular effects pertinent to the metastatic process including regulation of epithelial cell differentiation and proliferation, modulation of angiogenesis, synthesis of extra cellular matrix proteins (MMPs) and integrin receptors, and alterations in production of matrix degrading proteinases and their inhibitors.
