N-myc downstream regulated gene 1(NDRG1) v/as cloned from colon carcinoma cell line HT29-D in the course of inducing differentiation by Van Belzen in 1997. The gene can express in many tissues, such as kidney, heart, brain, bone muscle, placenta, lung, prostrate, small intestine and ovary. But in leucocyte, immunity organ and liver ,etc, its expression was low. The universality expression of NDRG1 indicated that this gene might have important role in the course of histiocytic growth and differentiation.Many scholars have researched the expression of NDRG1 in tumors. Kurdistani found that NDRG1 was low expressed in many tumor cell lines and carcinoma tissues, such as breast carcinoma, prostate carcinoma and colon carcinoma. Some scholars also proved that high expression of NDRG1 could inhibit tumor growth. Transfected NDRG1 cDNA into tumor cells can inhibit cell growth and xenograph growth in nude mice.But some researchers indicated that the expression of NDRG1 was high in many tumors. Cangul founded that in many cancers such as lung cancer, brain cancer, melanin tumor, liver cancer, prostate cancer and kidney cancer, the protein expression of NDRG1 was high than normal control group. And this is due to the severe hypoxia in cancer tissues. So, NDRG1 can be a marker of tissue hypoxia.Induction differentiation therapy is a new field in tumor therapy. Inducing differentiation refers to a reversible process of the malignant cancer cells towards the normal by exogenous and endogenous differentiation reagents.
