OBJECTIVE:To investigate the significance of LOH (loss of heterozygosity) events at specific chromosome regions in the tumorigenesis and progression of gastric adenocarcinoma in the Chinese population, to determinate the expression of candidate TSGs in these regions, and to identify the relevance of those TSGs' expression with clinicopathological features of gastric adenocarcinoma.METHODS:A panel of 9 polymorphic microsatellite makers on 3p14.2, 9p21, 16q22.1, 17p13.1, 18q21, and 21q21, including D3S1300, D3S1234, D9S1748, D16S496, D16S512, TP53, DCC, D18S46, and D21S1436, were used to investigate the LOH of specific chromosome regions respectively. PCR and PAGE were used to detect LOH of these MS markers in 38 gastric adenocarcinomas, and the relevance of LOH events with tumorigenesis and progression was analyzed. Expression of FHIT gene and E-cadherin gene was investigated by immunohistochemistry, and association of expression of these two genes with clinicopathological features and biological behavior of 38 gastric adenocarcinomas in this group was analyzed respectively.RESULTS:Nonrandom LOH events were observed for all of the MS markers investigated. LOH frequencies of D3S1300 and D3S1234 on 3pl4.2 were 30% (9/30) and 33.3% (9/27) repectively, and LOH of at least one MS marker was observed in 13 out of 28...
