Part One Isolation, purification and identification of CD34~+ human hematopoietic stem cells(HSC) from human umbilical cord blood (UCB).Objective: To establish the methods for isolation, purification and identification of CD34~+ HSC from human umbilical cord blood.Methods: UCB were obtained from fullterm deliveries collected after written informed consent in our hospital. The donors were healthy with both HBV and HCV negative. Low-density mononuclear cells were separated from UCB by density gradient centrifugation. CD34~+ HSC were further isolated from UCB samples by magnetic activating cell sorting (MACS) isolation. These sorted CD34~+ stem cells were directly used in the transplantation experiments or stored in liquid nitrogen. Use fluorescence-activated cell sorter (FACS) to detect the surface markers of CD34~+ HSC.Results: After density gradient centrifugation, the amount of MNC was nearly 1/10 of WBC in each portion of umbilical cord blood.
