Part â… STUDY ON THE MECHANISMS OF PACLITAXEL-INDUCED ACTIVATION-OF NUCLEAR FACTOR-κB IN HUMAN OVARIAN CANCER SKOV-3 CELL LINEObjective To investigate whether paclitaxel can induce activation of NF-κB in human ovarian cancer SKOV-3 cell line. To discuss the acting mechanism after paclitaxel-induced activation of NF-κB in SKOV-3 cells treated with paclitaxel.Methods SKOV-3 cells were treated with various concentrations of paclitaxel at various time intervals. Western blot assay was used to determine the nuclear p65 protein and cytoplasmic IκBa protein. RT-PCR assay was used to determine the mRNA of cIAP-1 anti-apoptosis gene. Cell apoptosis and cell cycle were determined by flow cytometry and cell growth was determined by MTT.Results The results of Western blot test reveal that there is a little P65 protein expressed in the nucleus of untreated cells and the IκBa protein is obviously...
