Research Of The Relationship Between HBV Infection And HLA Class I Expression On Host Lymphocytes

Objective: To evaluate the expression of HLA class I and HLA class II on peripheral blood lymphocytes in patients with chronic hepatitis B,hepatic cirrhosis and hepatocellular cancer. To set up method of RT-PCR for evaluating HLA-A mRNA in peripheral blood lymphocyte,To induce peripheral blood lymphocytes with IL-2 in such patients and carry out LDH releasing test.To study the role of the expression of HLA class I and HLA class II on peripheral blood lymphocytes in host celluar immunity in patients with chronic hepatitis B,hepatic cirrhosis and hepatocellular cancer.To provide a new train of thought for the mechanism of disorder of celluar immunity.Methods:1. To evaluate the expression of HLA class I and HLA class II on peripheral blood lymphocytesTo label lymphocytes by antibody: prepare four test tubes marked as HLA-I-Ctr, HLA-II-Ctr, to take 10 ul of isotype control immunoglubin into the tube of HLA-I, HLA-II.in the last,to take 100 ul anticoagulated blood into the tubes above.To adjust the instrument with Calibrite beads,to take the HLA-I-Ctr,HLA-II-Ctr as negative controL.To circle the region of lymphocytes in the 2-dimensional Dotplot picture of FSC and SSC,to collect 10~4 cells and evaluate their fluoresecence intensity of FITC which is amplificated as logarithm,to save data of light reflex.
