| Aim:The incidence of vascular dementia has been higher and higher in recent years.It has become an important common sanitation problem.According to my tutor's long-term clinical practical experiences,he created a prescription named KangNaoLing capsule which main function is promoting blood circulation and removing the blood stasis combined with nourishing kidney-jing and complement intellect.We received a satisfying curative effect by using this prescription on our patients in the clinical practice.In order to find out the effect mechanism of this prescription in curing vascular dementia,we stressed the study on apoptosis in neurocyte of vascular dementia with KangNaoLing capsule.Methods:The rat model of ischemia- reprefusion brain injury based on high-lipid food was used in this study. We detected the change of rats′learning and memory ability by Morris water maze experment,detected the lipids level and viscosity of blood , detected nitric oxide(NO), nitric oxide synthase(NOS),super-oxide dismutase (SOD),MDA of brain tissue.We principally observed the apoptosis in neurocyte, Intracellular free Ca2+ concentration,and the expression of p53,bcl-2 and caspase-3 gene.Result:KangNaoLing capsule can improve the learning and memory ability of rat model. It can also improve the free radical metabolism, reduce the level of blood lipids, decrease the blood viscosity, increase the level of NO and enhance the activity of NOS.We also found that KangNaoLing capsule can remarkably restrain apoptosis in of rat model.It can enhance the expression of bcl-2 gene, decrease the expression of p53 and caspase-3 gene, weaken calcium surcharge in neurocyte.Conclusion:KangNaoLing capsule is a good prescription for vascular dementia patients which is safe and effectual.Its effect mechanism maybe relate to restraining apoptosis in neurocyte, improving the free radical metabolism, reducing the level of blood lipids, decreasing the blood viscosity, increasing the level of NO and enhancing the activity of NOS.
