OBJECTS: To investigate the effects and mechanism of a selective COX-2 inhibitor NS-398 on the proliferation of human cervical carcinoma cell lines Hela.METHODS: The effect of NS-398 , carboplatin, and both of them on the proliferation of human cervical carcinoma cell lines(Hela) was performed by MTT growth assay , and the data were analyzed using SPSS general linear models procedure to determine the IC50 of NS-398. After the cells were treated with the concentration of NS-398, the apoptosis assay and the cell cycle distribution were analyzed by Flow cytometry. 2-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix associated laser desorption /ionization time-of-flight mass spectrometry (MALDI-TOF-MS), coupled with bio-informatics were carried out to find the differently expressed proteins in the human cervical carcinoma cells treated with NS-398.RESULTS: 1)The cervical carcinoma cells were divided into four groups. They were the control group, NS-398 group, carboplatin group and cooperation group .
